The BD Rhapsody™ Single-Cell Analysis System was introduced in 2017 as a microwell-based Single-Cell Analysis system to generate NGS libraries from thousands of single cells.
The simple and robust workflow has enabled scientists around the world to decipher complex biological systems in great detail without the risk to lose precious samples to wetting or clogging errors.
With the launch of the BD Rhapsody™ HT Xpress it is now possible to process hundreds of thousands of single cells in one run.
The BD Rhapsody™ 8-Lane Cartridge was specifically designed for the new BD Rhapsody™ HT Xpress, offering 8 independent lanes with ~270,000 microwells per lane. Analyze your samples with different assays or assay combinations in the 8 lanes of the cartridge.
What if you do not need all lanes simultaneously? Load samples into unused lanes on another day!
The t-SNE plots below show correlations of an experiment in which single lanes of an eight-lane cartridge were loaded on day one, day seven, and after four months of storage. No batch effect was observed, the correlation of gene expression was high between the single lane and the 8-lane cartridge, and lane-to-lane variability was minimal. The results may vary based on cell type and isolation method.
Lanes 1 and 2 of a BD Rhapsody™ 8-lane Cartridge were loaded with 20,000 cells consisting of Jurkat and Ramos cells at a 1:1 ratio and on a single-lane cartridge as a control. Targeted and SMK libraries were prepared from subsampled beads at 3,500 cells using the BD Rhapsody™ Targeted kit with the BD® Immune Response Panel (HS) and BD® Sample Multiplexing Kit (SMK).
Lanes 3 and 4 of a BD Rhapsody™ 8-lane Cartridge were loaded with 20,000 cells from a single PBMC donor at seven days storage of the partially used cartridge and on a single-lane cartridge as a control. WTA and AbSeq (10-plex) libraries were prepared from subsampled beads at 3,500 cells on day 7 storage using the BD Rhapsody™ WTA Amplification Kit.
Lanes 7 and 8 of a BD Rhapsody™ 8-lane Cartridge were loaded with 20,000 cells consisting of Jurkat and Ramos cells at a 1:1 ratio at four months of storage of the partially used cartridge and on a single-lane cartridge as a control. Targeted and SMK libraries were prepared from subsampled beads at 3,500 cells as on day one.
